Retatrutide (LY3437943) is a single polypeptide conjugated to a fatty diacid moiety that activates GIP, GLP-1, and glucagon receptors. The triple agonism provides complementary metabolic benefits: GLP-1 reduces appetite and slows gastric emptying, GIP enhances insulin sensitivity and may promote fat storage efficiency, and glucagon receptor activation increases hepatic fat oxidation, energy expenditure, and thermogenesis. Reconstitution steps from the dataset
- Clean. Remove flip-top cap and swab rubber stopper with alcohol.
- Draw water. Draw 2mL of bacteriostatic water into syringe.
- Inject water. Insert needle through stopper and slowly inject water down the side of the vial. Do not spray directly onto powder.
- Dissolve. Gently swirl (do not shake) until fully dissolved. Solution should be clear.
- Label. Label vial with date and concentration (5mg/mL). Refrigerate immediately.
- Dose. For 1mg dose: draw 0.2mL (20 units). For 2mg dose: draw 0.4mL (40 units). Inject subcutaneously once weekly.
