MOTS-c

Mitochondrial Open Reading Frame of the 12S rRNA-c

MOTS-c is a mitochondrial-derived peptide (MDP) encoded within the mitochondrial genome's 12S rRNA gene. It is the first mitochondrial-encoded peptide shown in published research to regulate nuclear gene expression — making it a signaling molecule between mitochondria and the cell nucleus. Researchers study MOTS-c for its documented effects on metabolic homeostasis, exercise physiology, and cellular energy production.

Key fact

First mitochondrial-derived peptide shown to regulate nuclear gene expression; functions as an exercise mimetic via AMPK activation

Mechanism

MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA Type-c) is a 16-amino acid peptide encoded by the mitochondrial genome (12S rRNA gene). It is the first mitochondrial-derived peptide (MDP) shown to regulate nuclear gene expression. In response to metabolic stress or exercise, MOTS-c translocates to the nucleus where it regulates antioxidant response element (ARE) genes, activates AMPK signaling, enhances skeletal muscle glucose uptake via GLUT4 translocation, increases fatty acid oxidation, and improves mitochondrial respiration. Functions as an exercise mimetic by activating exercise-responsive gene programs.

Evidence note

Experimental — Highly promising preclinical data. Limited human data (small studies, mostly observational correlations). The mitochondrial origin and exercise-mimetic mechanism are scientifically novel and well-published, but clinical translation is early.

Male in my 40s, have played around with different peptides, seem to be very sensitive, but also find them to be very effective. Did a bit of an impulse buy of some MOTS-C with an order . One of the "reputable" research places, so felt ok there. Started off this morning w/ 2mg.

I almost dropped MOTS after like 3 weeks because it felt like nothing. Was running it with Tirz (5mg weekly) and added MOTS at 10mg twice a week. First few weeks were basically nothing… no energy change, no appetite difference. Then around week 4 I realized I wasn’t getting

I have a love hate relationship with coffee. I’ve known for years that it has dehydrated me, created joint pain, neck pain and worn out my adrenals. When I started a course of Mots c, the energy and vitality inspired me to start weening off caffeine. I just don’t tolerate it

FAQ

Is MOTS-c naturally produced?

Yes. MOTS-c is encoded in the mitochondrial genome and naturally produced by cells. Levels increase with exercise and decline with age.

What is an exercise mimetic?

A compound that activates some of the same cellular pathways as physical exercise — particularly AMPK activation and metabolic adaptation. MOTS-c is the most studied mitochondrial exercise mimetic.

Why is it considered a 2026 breakout?

Growing mainstream media coverage, accelerating research publication rate, and endorsement by prominent longevity researchers have driven rapid demand growth.
