Kisspeptin-10
Hypothalamic GnRH-Activating Decapeptide
Kisspeptin-10 is the bioactive fragment of the kisspeptin family — neuropeptides that act on the GPR54 receptor to control GnRH (gonadotropin-releasing hormone) secretion from the hypothalamus. Discovered in the early 2000s, kisspeptin signaling was identified as the master regulator of the reproductive hormone axis. Researchers study it because it sits at the very top of the HPG cascade, upstream of GnRH, LH, FSH, and all downstream sex hormones.

Key fact
Master upstream regulator of the HPG axis. Potent LH/testosterone stimulator via GPR54/GnRH pathway. Published in JCEM and Nature. More physiological than hCG.
Mechanism
Kisspeptin-10 is a decapeptide (C-terminal 10 amino acids of kisspeptin-54) that binds KISS1R (GPR54) on GnRH neurons in the hypothalamus. This receptor activation triggers GnRH release, which stimulates anterior pituitary gonadotrophs to secrete LH and FSH. In men, kisspeptin-10 boluses potently evoke LH secretion, and continuous infusion increases testosterone, LH pulse frequency, and pulse size. The kisspeptin-GnRH-gonadotropin axis is the master regulator of pubertal onset and reproductive function. Shows sexual dimorphism (stronger LH response in men during follicular equivalent).
Evidence note
Emerging — Human clinical trials demonstrating potent LH and testosterone stimulation. Published in top-tier journals (JCEM, Nature). Active clinical research for IVF and reproductive disorders. But not yet approved for any indication, and community experience is limited.

FAQ
How is kisspeptin-10 typically dosed?
Clinical trials used 1-10mcg/kg IV bolus. Community protocols typically use 100-500mcg subcutaneously daily or every other day. Dosing data is limited compared to more established peptides — most protocols are extrapolated from clinical research.
Can kisspeptin-10 be used for PCT (post-cycle therapy)?
Kisspeptin-10 is being studied as a potential HPG axis restoration tool. It sits at the very top of the hormonal cascade — above GnRH — making it theoretically useful for restarting suppressed hormone production. However, its short half-life requires frequent dosing, and community experience with this application is limited.
How does kisspeptin-10 compare to hCG for testosterone support?
Kisspeptin works upstream — it stimulates GnRH which then triggers LH/FSH release, working through the body's natural HPG cascade. hCG mimics LH directly at the gonadal level, bypassing the hypothalamus and pituitary. Kisspeptin is considered a more physiological approach since it preserves the entire hormonal feedback loop.
